application | dutasteride dihydrogen is also called dihydrotasteride, which is an intermediate for synthesizing dutasteride. Dutamide is mainly used clinically for prostate enlargement, male pattern alopecia, seborrheic alopecia, and hereditary alopecia (women and children should not take it). |
preparation | 16.65g 3-keto -4-aza-5 α-androst-17 β-carboxylic acid methyl ester was dissolved in 250 ml of dichloromethane, and 18.95 ml of boron tribromide was added dropwise under the cooling of room temperature water (25 ℃). Continue to stir at room temperature for 20 minutes to obtain a clear brown solution. 19.5 ml of 2,5-bis (trifluoromethyl) aniline was added to the above solution under cooling with room temperature water (25 ℃). Heat the reaction to 50 ℃ and stir overnight. After the reaction solution is cooled to room temperature, 50 ml of water is slowly added, and the generated hydrogen bromide gas is introduced into the Na2CO3 aqueous solution. Continue to add 200 ml of water until all solids dissolve and get clear two phases. The isolated lower dichloromethane organic phase was dried with anhydrous Na2SO4 after washing with 200 ml of water and 200 ml of saturated salt. After 10 grams of silica gel filtration, reduced pressure concentration to 60 ml. Slowly add 100 ml of petroleum ether dropwise to the above concentrated solution. The product is precipitated with tasteride dihydrogen in a light yellow solid degree. After filtration and petroleum ether washing, the product is dried to a constant weight of 24.65g(93%) in the air. |
biological activity | Dihydro Dutasteride are metabolites of Dutasteride. Dutasteride are inhibitors of 5α reductase isoenzymes. |
